بررسی تنوع ژنتیکی ژنوتیپ های گندم دیم با استفاده از نشانگرهایRAPD و صفات مورفولوژیکی

چکیده

تنوع ژنتیکی در بین ژنوتیپ‌های گندم دیم برای اهداف بهنژادی بسیار مهم است، زیرا ژرم پلاسم یکنواخت ممکن است در معرض تنش‎های زیستی و غیر زیستی قرار گیرد. در این تحقیق تنوع ژنتیکی 25 ژنوتیپ گندم دیم با استفاده از صفات مورفولوژیکی و 20 آغازگر RAPD  بررسی شدند. ژنوتیپ­ها از نظر اغلب صفات مورفولوژیکی اختلاف معنی­دار داشتندکه بیانگر وجود تنوع ژنتیکی بین آن‎ها است. بر اساس داده‎های مولکولی، در مجموع 197 نوار متمایز در 25 ژنوتیپ بدست آمد. میانگین تعداد نشانگر به‎ازای هر آغازگر 8/9 بود. از تعداد کل نوارها، 160 نوار چند شکل بودند. با توجه به ماتریس تشابه ژنوتیپ­ها و داده‌های مولکولی و مورفولوژیکی بر اساس فاصله اقلیدسی آن‎ها، دامنه تشابه ژنوتیپ‌ها به ترتیب بین 56/0 تا 83/0 و 92/1 تا 69/108 متغیر بود. بیشترین میزان شباهت بر اساس نشانگر RAPD و صفات مورفولوژیکی به ترتیب مربوط به ژنوتیپ‌های مارون با گهر(83%) و کوهدشت با Pigo (92/1) بود. نشانگر RAPD و صفات مورفولوژیکی توانستند ژنوتیپ­های تیپ بهاره، زمستانه و هم‎چنین ژنوتیپ­های با عملکرد بالا (Nestor, TV2, Katila-11 و GHK) را به‎خوبی از هم تفکیک نمایند، ولی در تفکیک ژنوتیپ‌های گندم نان و دوروم نشانگرها متفاوت بودند. بین ماتریس تشابه داده‌های مولکولی و صفات موفورلوژیک همبستگی منفی ولی غیرمعنی‌دار وجود داشت.

چکیده [English]

 Diversity is very important for the breeding objectives, since a narrow genetic base of germplasm is very vulnerable to biotic and abiotic stresses. In this study, genetic diversity of 25 rainfed wheat genotypes were assessed using morphological traits as well as 20 primers of RAPD markers. There were significant differences among genotypes for the most traits, indicating high genetic variations among wheat genotypes. Based on the molecular data, 197 bands were detected within 25 genotypes and 160 bands were polymorphs. Average mean number of bands per primer was 9.8. According to cluster analysis of similarity matrix of molecular and morphological data based on euclidean distances, similarities ranged between 0.56-0.83 and 1.92-108.96, respectively. Based on molecular data and morphological traits, the highest similarities were belonge to Gahar and Maron (0.83), and Kuhdasht and Pigo (1.92) varieties, respectively. The results showed that RAPD markers and morphological traits could distinct spring and winter wheat genotypes having high grain yield (Nestor, TV2, Katila-11 and GHK). Distinction of durum and bread wheat genotypes did not follow the same pattern. There was no significant correlation between two similarity matrices of molecular and morphological data.
